A genetic linkage map of Theobroma cacao L. based on isozymes, RFLPs, and RAPDs markers

A linkage map of cacao genome have been constructed using isoenzymes, RFLPOs, and RAPDs markers. A genomic library and cDNA library were constructed from germinated seeds of an Amelonado clone. The screening of probes was made on 5 genotypes, using 5 restriction enzymes. Among 587 tested probes, 52% of cDNA and 44% of genomic probes revealed polymorphic patterns. A population of 100 individuals derived from a cross between two heterozygous genotypes has been used. Segregation analyses were performed with JoinMap programm. The map comprised 193 loci: 5 isoenzymes, 101 cDNA/RFLPs, 4 loci from genes of known fonction, 55 genomic DNA/RFLPs and 28 RAPDs. Ten linkage groups, which putatively correspond to the 10 gametic chromosomes of cacao, have been identified. The map covers a total length of 759 cM with a 3,9 cM average distance between two markers. A small fraction (9%) of the markers deviated significantly from the expected mendelian ratios
