During the present mission, mature embryons have been isolated from 300 Kopyor (Macapuno) coconuts. The sampling protocol and the schedule of forthcoming operations have been fixed, in order to obtain viable plantlets in nursery within 5 months (November 1991). The first results concerning contaminations and oxidations after 2 weeks in culture are mostly encouraging, showing an average rate of success reaching 96.5%. The definitive protocol includes: disinfection of dehusked coconuts with ethanol 96° (1 minute), then NaOCl 2.5% (5 minutes), disinfection of isolated embryos with NaOCl 1.0% (20 minutes) followed by a rinsage (5 passages) in sterile water. The presence of an IRHO expert is requested for the section of the haustorium after 3 months in culture (1st September 1991).
