Development and integration of microsatellite markers into a molecular linkage cocoa map

Microsatellites or simple sequence repeat (SSR) markers have been widly used in many crop species due to their abundance, high degree of polymorphism, locus specificity, reproductibility and codominance heredity. Two hundred fifty newly developed microsatellites were produced from a genomic library enriched in SSR and integrated into an existing high density molecular linkage cocoa map with 424 markers including 20 microsatellites (Risterucci et al, 2000) The same mapping population resulting from a cross between two heterozygous genotypes, a Forastero and a Trinitario (hybrid between Forastero and Criollo) was used for the linkage analysis. The new microsatellite markers were assigned to ten different linkage groups that correspond to the ten gametic chromosomes of cocoa allowing a widespread distribution throughtout the cocoa genome. This map will be a useful tool to identify the most important regions involved in the variation of the traits of interest and to develop Marker Assisted Selection. (Texte intégral)
