Characterization of the first Coffea canephora BAC library [S04-17]

A BAC library of the heterozygous diploid genotype of C. canephora 126 was constructed starting from young leaf plant material. The BAC library is constituted of 55,296 clones with an average insert size of 135 Kb which represent a coverage of 9 haploid genome equivalents. The validation and characterization of the BAC library are underway to estimate the extent of contamination by organelle DNA (chloroplastic and mitochondrial). The use of single-copy probes will enable to experimentally validate the genome coverage of the BAC library. The BAC library represents a valuable and central tool to investigate the existence of linkage disequilibrium in wild populations of C. canephora descended from a small number of mother-trees. The BAC library will also be used to physically delimit QTLs for important agronomic traits and to characterise genes controlling sucrose content during coffee bean development. (Texte intégral)
