Caractérisation moléculaire des champignons destructeurs des bois tropicaux

Five brown-rot fungi (Tyromyces palustris, Gloeophyllum trabeum, Antrodia sp., Poria placenta, Coniophora puteana) and three white-rot fungi (Coriolus ve.rsicolor, Pycnoporus sanguineus, Lentinus squarrosulus) registered as representative of international standards of wood durability tests have been genetically characterized through PCR/RFLP techniques. The Internal Transcribed Spacer between 18s and 25s rRNA has been amplified using universal primers. The amplified fragments were compared by using ten restriction enzymes. The analysis of RFLP gel electrophoresis allowed to clearly discriminate all the strains from each others. These RFLP analyses constitute a range of reference profiles to further analyze the genetic diversity of the 150 wood-degrading fungi strains currently in collection in the laboratory.
