Research on pathogenic variation of Ralstonia solanacearum strain Po82 and its genome

R. solanacearum, as a species complex, can be divided into four phylotypes on the basis of phylotype subclassification system proposed by Fegan and Prior. In our phylogenetic research, Po82, a potato bv1 strain from Mexico, was resolved into phylotype II sequevar 4, in a group with phylotype II/4NPB and banana Moko disease-causing strains. It was identified as Moko strains' sequevar 4 (MLG25) according to Mmx-PCR. Additionally, pathogenicity test revealed that Po82 possess pathogenic traits of both NPB and Moko strains thereby proving that it was a pathogenicity variation strain. The genome sequence of the Po82 genome was determined by using a massively parallel pyrosequencing technology (Roche 454 GS FLX) .The genome size of Po82 is 5.34 Mb, with an average GC content of 66.7%. Compared to seven sequenced R. solanacearum strains, 465 strain-unique genes were identified within the genome of strain Po82, most of which encode hypothetical and conserved hypothetical proteins. Furthermore, 48 putative genomic islands were predicted within Po82 genome.
