Comparative assessment of molecular tests for the detection of Xanthomonas citri pv. citri in the framework of European H2020 project Valitest

Early diagnosis and rapid management are crucial to reduce the risk of entry, spread and establishment of plant pests and to limit their damage. VALITEST* (an EU funded research project (https://www.valitest.eu)) coordinated the implementation of two rounds of test performance study (TPS) for prioritized pests in a range of matrices and for a range of diagnostic technology related platforms (both laboratory and on site-based). The main objective of this work was to produce validation data for the selected tests where no or limited validation data were available. ANSES Plant Health Laboratory – Unit for Tropical Pests and Diseases, located in Reunion Island organized the TPS for the detection of Xanthomonas citri pv. citri, the causal agent of Asiatic citrus canker. Eighteen participants from EU countries, Australia and New-Zealand participated in this TPS. Thirteen different molecular tests (based on conventional PCR, real-time PCR and LAMP (Loop-mediated isothermal amplification)) were evaluated in the framework of this TPS, through a panel of 24 Citrus samples composed of target samples, non-target samples, and serial dilutions of target samples. Results of the TPS allowed to distinguish significant differences between tests mostly in terms of analytical specificity, analytical sensitivity, diagnostic sensitivity & specificity, and reproducibility. Results were used to contribute to the revision of the EPPO protocol PM7/44 concerning the detection of Xanthomonas citri pv. citri and Xanthomonas citri pv. aurantifolii. *Grant agreement n°773139
